In most centers the ER analysis is performed on surgically removed tumor specimens. However, in many patients with inoperable or recurrent carcinoma, surgical specimens for ER determinations are not readily available, and after preoperative irradiation ER determination is unreliable as the primary tumor often undergoes advanced degenerative changes or totally disappears [6]. In such cases ER determination on fine needle aspirates could be a method of choice. Two earlier reports based on a small number of patients [4, 5] indicated that reproducible ER levels may be obtained from analysis of fine needle aspirates from breast carcinoma.

Quality Control. Systematically, a cytosol reconstituted from a lyophilized rabbit uterus primed with estradiol before killing is introduced in the assay. Results Sample Characteristics Size and Weight. 5-2 mm in diameter. Several small fragments are obtained in soft tumors. Samples containing well-defined fragments generally weigh more than 20 mg. Such samples are obtained in 90% of cases. When the tumor is particularly soft, debris can be obtained; this pertains to 9% of cases. Complete failure to obtain a tissue sample is generally due to technical problems and is very rare (1% of cases).

As shown in Fig. 2, this leads to point 2 appearing as an unacceptable outlier. Recalculation after rejection of both points 1 and 2 produces a reasonable linear plot with some scatter of points (Fig. 3). However, a hand-drawn curve (which mayor may not accurately represent a multicomponent system) would appear to give a better fit (Fig. 4). In such a case a full analysis requires goodness of fit data for several possible models rather than arbitrary rejection of data.
